This innovative sample preparation method makes use of optimized deglycosylation conditions and reagents to enable a fast release. The novel reagents included were purposefully designed to provide the benefits of both sensitive fluorescence detection and superior matching signal intensity for mass detection. Before liquid chromatography analyses, sample clean-up is accomplished using solid-phase extraction with a GlycoWorks HILIC µElution Plate.

This refill kit includes materials to process 24 samples in a 3x8 format and comes with GlycoWorks Deglycosylation Module and the GlycoWorks Labeling Module shipped together to supplement other kits or as reagents for GlycoWorks RapiFluor-MS N-Glycan Kit- 96 Sample. Additionally, this kit includes GlycoWorks Deglycosylation Module (1), 0.035 ml/vial Rapid PNGaseF Enzyme (1), 0.25 ml/vial GlycoWorks Rapid Buffer (3), 3 vials of 3 mg/vial RapiGest SF Surfactant, GlycoWorks RapiFluor-MS Labeling Module (3), 9 mg/vial RapiFluor-MS Reagent (3), 1 ml/vial Reagent Solvent (Anhydrous DMF). All lab equipment is manufactured to industry and regulatory standards, with the appropriate vessels for each substance being used for packaging.

How Should I Equilibrate My Columns Before Use With RapiFluor-MS?

Before getting started with Glycan analysis, a newly received Glycan BEH Amide Column should be flushed in 60% acetonitrile: 40% aqueous for 50 column volumes. Equilibrate with 20 column volumes of initial mobile phase conditions before making the first injection. Between injections, equilibrate with 8-10 column volumes. Failure to appropriately follow these steps to equilibrate the column may lead to drifting retention times.
