Based on Waters’ novel ethylene bridged hybrid (BEH) particle, XBridge BEH Amide Columns utilize a chemically stable, trifunctionally bonded amide phase to enable a new level of stability and versatility for HILIC separations. With the same selectivity as ACQUITY UPLC BEH Amide columns, scientists can easily transfer separations between HPLC and UPLC lab equipment as well.

XBridge BEH Amide Columns were designed specifically to overcome common challenges in retaining and separating extremely polar compounds and metabolites that are too polar to be retained by reversed-phase chromatography. The columns facilitate the use of a wide range of mobile phase pH (2 – 11) in order to facilitate this exceptional retention of polar analytes that spans a wide range of polarity, structural moiety, and pKa.

Along with enhanced retention of polar compounds, XBridge BEH Amide columns offer an increased mass spectrometry response, direct compatibility with sample preparation eluates like PPT, LLE, and SPE, and orthogonal selectivity when compared to reversed-phase materials.

XBridge BEH Amide Columns are well suited to a range of applications, including the analysis of carbohydrates. The 3.5 µm particle size enables high resolution, high-speed analysis of carbohydrates in complex sample matrixes while maintaining or improving chromatographic resolution. In addition to this, increased chemical stability allows for the use of high pH and high temperature to collapse anomers without the loss of reducing sugars. Unlike amine-based columns for this application, these columns are not susceptible to Schiff base formation, allowing for improved quantitation accuracy Before use in carbohydrate analysis, scientists can benchmark separations using HILIC QC Reference Material.

What Are the Operating Limits For XBridge BEH Amide Columns?

The recommended pH limit for XBridge BEH Amide Columns includes a wide range of 2 – 11. These columns can tolerate pressures of up to 6,000 psi (400 bar or 40 Mpa). However, pressures greater than 4,000 to 5,000 psi should be avoided when possible to maximize column and system lifetimes. XBridge BEH Amide Columns have a recommended temperature limit of 90 degrees C in order to enhance selectivity, lower solvent viscosity, and increase mass transfer rates. However, temperatures above ambient will have a negative effect on lifetime, which can vary based on the pH and bugger conditions used.
