RapiGest™ SF, is an innovative anionic surfactant that drastically improves in-solution protein digestion in terms of speed and peptide recovery.  It is especially valuable in protein sample preparation for mass spectrometry (MS) analysis by avoiding high background noise often seen from other common surfactants. 
RapiGest™ SF is available in a variety of formats.  This part contains one, 50 mg vial for large scale digestions. Add RapiGest™ SF to your proteolytic digestion, (using trypsin or other enzymes) to enhance digestion speed and increase peptide recovery.

RapiGest SF, 50 mg, 1/pk

Originally created as an enzyme-friendly surfactant meant to assist in-solution protein digestion, RapiGest SF is an innovative evolution of the product that helps to improve protein solubility during preparation of samples. As a Waters patented reagent, this lab equipment, RapiGest SF is used to enhance in-gel and in-solution enzymatic digestions of proteins. It is particularly beneficial when preparing protein samples for mass spectrometry (MS) analysis because of the ability to avoid high levels of background noise that results from other common surfactants.

RapiGest SF has also been shown to be useful as a method to solubilize glycoproteins before deglycosylation with enzymes like PNGase F. This process is also beneficial as proteins are made more vulnerable to enzymatic cleavage while avoiding inhibition of enzymatic activity, and remaining heat stable to support high-temperature digestions.

Use of RapiGest SF will not modify peptides or suppress endoprotease activity, making it compatible with a wide range of enzymes, including Trypsin, PNGase F, Lys-C, Arg-C, Asp-N, Glu-C. It can also be easily removed after use to allow for MALDI-TOF MS, LC, or LC/MS analyses of already digested samples.

This surfactant works through mild protein denaturation, which opens protein structure and exposes the proteolytic sites to enzymatic cleavage. These enzymes are left more resistant than common proteins, allowing them to remain active in RapiGest SF solutions. The hydrolysis in acidic conditions that RapiGest SF undergoes allows it to be used to remove surfactant from solutions when necessary.

Can RapiGest SF be removed after digestion?

In order to remove the surfactant after digestion, no special detergent steps like dialysis are required before analysis occurs. The enzymatic digestions are generally acidified with acids like formic acid, trifluoroacetic acid (TFA), or hydrochloric acid (HCI) in order to degrade RapiGest SF before any further analysis is performed.

What acid pH level can this be performed with?

It is recommended this is performed with an acid of a pH less than 2. Further analyses like MALDI-TOF MS, LC, or LC/MS analysis can then be performed on the digested samples.
