The MassPREP Protein Digestion Standards are useful in a variety of applications that include performance evaluation of HPLC columns, high-performance liquid chromatography, and mass spectrometry instrumentation used for the analysis of this class of compounds. The MassPREP E. coli Digest Standard is intended to provide a complex biological matrix for producing samples used to train new users of the Waters Protein Expression System. The MassPREP E. coli Digest Standard was prepared by digesting a purified E coli. Cytosolic protein fraction with sequencing grade trypsin. Its mixture does not contain any undigested protein, trypsin, or very hydrophilic components such as salts.
The MassPREP E. coli Digest Standard should be stored at room temperature and has a shelf life of up to 60 months. Typically the lyophilized powder within the MassPREP E. coli Digest Standard is stable at room temperature until the expiration date on the label or one year from the purchase date. Upon reconstitution, its shelf life is just about a week, although stability can be extended by freezing samples at -20 degrees.
Typically one vial of The MassPREP E. coli Digest Standard will be required for executing the Protein Expression System training module using Nanoscale (75µm) HPLC. To prepare the MassPREP E. coli Digest Standard, resuspend lyophilized powder in 250 µL of 0.1% formic acid. Mix the capped vial by vortexing to ensure all material is resuspended.

The lyophilized powder in the first vial should be resuspending in 50µL of 0.1% formic acid. Using a pipette, you can transfer the solution created in the first vial to the second vial and use it for resuspending the remaining lyophilized powder.
