Waters Ammonium Formate Solution – Glycan Analysis
With the introduction of the reagent RapiFluor-MS, Waters has become a leader in innovation for biotherapeutic analysis, reimagining the workflow for released N-glycan analysis. Waters knows that characterizing and monitoring the glycan population of a biotherapeutic protein has continued to present technical and logistical challenges across the industry. We continue to drive technological advancements in pursuit of improving released glycan analysis. By combining the same preparation and analytical technologies, the Glycoworks system has provided analysts with smart workflows, rapid and streamlined sample preparation, and unrivaled FLR and MS response. This allows for a reduced time from sample to results, with sample cleanup able to be completed in as little as 30 minutes.
Part of this system, Waters Ammonium Formate Solution – Glycan Analysis is a concentrated version of the 50 mM Ammonium Formate buffer that is recommended for use with the Biopharm/Glycan system. It is also compatible with the UPLC H-class and lab equipment such as the ACQUITY UPLC Glycan BEH Amide Column, 130Å, 1.7 µm, 2.1 mm X 150 mm, 1K - 150K, 1/pk. The buffer contains 10 mL of 5050 mM Ammonium Formate in a water with 3.8% Formic acid matrix. The use of the buffer allows the scientist to directly pour the concentrate into a 1L bottle of LCMS grade waters, resulting in the correct concentration of 50 mM at a pH of 4.4.
How Should Waters’ Ammonium Formate Solution be Stored?
Initial storage is best done at room temperature. When longer than one month of storage is needed post solubilization, room temperature is no longer recommended. When not in use, it should then be stored in the fridge. Adequate time should be given for the solution to return to room temperature before use. When smaller volumes are needed, one could use 1 mL at a time in order to add up to an eventual 100 mL of water. Recommended storage temperature in the refrigerator is 5C +/-3C. The recommended shelf life of the solution is 12 months.
