The aim of this course is to study the fragmentation of positively and negatively charged even-electron ions of small molecules during in-source CID or MS/MS in triple quadrupole, ion trap, and quadrupole time-of-flight hybrid systems. The emphasis is on spectrum interpretation rather than on fragmentation mechanisms. In addition to lectures and ample time for questions and discussion, the course provides exercises in order to practice spectral interpretation.
Topics Covered
Information from mass spectra: Average and monoisotopic mass, mass defects, double-bond equivalents, the nitrogen rule, isotope peaks, adduct ions and adduct-bound dimers
Fragmentation techniques: Collision-induced dissociation (CID), In-source CID, Triple quadrupole, ion trap and quadrupole-time-of-flight (QTof) hybrid instruments.
General aspects of fragmentation: Basic fragmentation mechanisms as well as the ability to discuss fragmentation as charge-site-directed process are investigated.
Fragmentation of even-electron positive ions 1: Oxygen compounds -1: Alcohols, ethers, glycosides, aldehydes, ketones, flavonoids, and steroids.
Fragmentation of even-electron positive ions 2: Oxygen and sulphur compounds -2: Carboxylic acids, esters, triacylglycerols, as well as various sulfur compounds, e.g., sulfonamide antibiotics.
Fragmentation of even-electron positive ions 3: Nitrogen compounds -1: Amines, cyclic amines, quaternary ammonium compounds, and aromatic nitrogen compounds like triazines.
Fragmentation of even-electron positive ions 4: Nitrogen compounds -2: Amides, carbamates, organophosphorous compounds, halogenides, nitro-compounds, nitrosamines, and some aromatic heterocyclic ring compounds.
Strategies for confirmation and identification using LC/MS/MS.
