Achieve quick, clean, and complete protein digestion

Protein digestions for biopharmaceutical peptide mapping or bottom-up proteomics are plagued by long digestion times and complex performance trade-offs. Balancing speed and digestion completeness while maintaining high reproducibility, low artifacts, and complete sequence coverage is also a constant challenge. Moreover, productivity during data analysis is often hampered by messy maps with various non-tryptic peaks (missed cleavages, autolysis peaks, non-specific cleavages, and unmatched peaks). Those numerous small, unexpected peaks require time consuming identification, consequently slowing down your critical work.

RapiZyme Trypsin, Mass Spectrometry (MS) Grade enables uncompromising speed and digestion cleanliness through its inordinately high autolysis resistance, high purity, and improved activity. By virtually eliminating the autolysis that plagues even the most advanced trypsin on the market, RapiZyme Trypsin can be used at comparatively high concentrations to achieve faster digestions without resorting to high temperature or sacrificing digestion completeness. Consequently, improving sample turnaround time, lab operations, and most importantly, making critical decisions confidently and quickly.

How RapiZyme works

RapiZyme Trypsin is a chemically optimized recombinant trypsin. Because it is not animal derived, it is free of contaminating chymotrypsin, does not require treatment with L-(tosylamido-2-phenyl) ethyl chloromethyl ketone (TPCK) inhibitors, and thus is free from non-specific digestion associated challenges.

Rapid Digestions

Extensive and homogenous methylation confers RapiZyme Trypsin with inordinately high autolysis resistance. This enables use of RapiZyme Trypsin at high E:P ratios (e.g., 1:5) to drive complete digestions in just 30-minutes, without resorting to higher temperature. Using Remicade® as a case study, RapiZyme Trypsin delivers <0.1% autolysis peaks and <1% missed cleavage (measured as % of total TIC response). An industry leading competitive trypsin delivers 6–7% autolysis peaks and 4–5% missed cleavage under the same conditions.

Clean baselines

Critical decisions are made by recognizing the smallest changes in a map. Regulatory agencies demand identification of every peak. Unmatched peaks, artifact peaks, autolysis peaks, and missed cleavage peaks are all time-consuming and unnecessary noises. They require extra scientific review and analysis, causing software processing times to shoot through the roof when non-tryptic peaks, or autolysis peaks from an in-silico model of trypsin digesting itself are included.

With RapiZyme Trypsin, you can proceed quickly and confidently with exquisite baselines free from noise and artifact peaks. All with a 30-minute digestion.

Reproducibility and purity – what’s in your trypsin?

RapiZyme Trypsin, homogenously methylated, recombinant (non-animal derived) trypsin is free of contaminating chymotrypsin. Further it is lyophilized with a LC-MS compatible excipient to make reconstitution easy and predictable and improve recovery from the vial.

The result is exceptional reproducibility both batch-to-batch and digestion-to-digestion.

Flexible to use

RapiZyme Trypsin can do everything you expect any trypsin (in solution or immobilized) to do and much more. With RapiZyme Trypsin, you can:

  • Eliminate desalt & enhance consistency for MAM, PTM monitoring, bioprocessing and proteomics applications. RapiZyme trypsin maintains high activity with modest amounts of guanidine HCl
    • Dilute guanidine HCl to 0.5–0.7M and digest in 2 hours at a 1:5 enzyme:protein ratio (pH 7.5)
  • Control artifacts for any application when overnight digests are preferred without compromising digestion efficiency
    • Digest overnight with a 1:100 enzyme:protein ratio at room temperature and pH 6.5
  • Accelerate traditional 1:20 enzyme:protein ratio digests to 1–3 hours to afford additional flexibility
  • Heat and eliminate the need for immobilized enzymes for simpler or easier to digest proteins with thermally stable RapiZyme
    • Digest with a 1:10 enzyme:protein ratio for 1 hour at 70 °C

Offers and Discounts

Back To Top Back To Top

RapiZyme Trypsin, Mass Spectrometry (MS) Grade, redefines protein digestion with an unparalleled balance of speed, cleanliness, and efficiency, making it an essential tool for biopharmaceutical peptide mapping and bottom-up proteomics. Designed to overcome the common challenges of long digestion times, complex trade-offs, and unpredictable results, RapiZyme Trypsin offers a reliable solution that accelerates workflows, enhances reproducibility, and eliminates time-consuming artifacts.

Protein digestions have traditionally struggled to maintain high sequence coverage, low artifacts, and reproducibility without sacrificing speed or cleanliness. RapiZyme Trypsin stands out by delivering rapid, clean, and complete digestions, thanks to its extensive and homogenous methylation, which confers inordinately high autolysis resistance. This innovative feature allows for the use of high enzyme-to-protein (E:P) ratios, enabling complete digestions in as little as 30 minutes without resorting to elevated temperatures or compromising digestion quality. For example, under a 1:5 E:P ratio, RapiZyme Trypsin achieves less than 0.1% autolysis peaks and under 1% missed cleavage, a significant improvement over industry-leading competitor enzymes that produce 6–7% autolysis peaks and 4–5% missed cleavages under the same conditions.

Clean baselines are critical for confident decision-making in regulatory environments, where every peak must be identified with precision. RapiZyme Trypsin eliminates noise from unmatched peaks, autolysis artifacts, and non-specific cleavages, ensuring your data maps are free from unnecessary distractions. By significantly reducing processing times and scientific review effort, RapiZyme Trypsin enhances productivity and speeds up analysis, allowing you to focus on the insights that matter most.

Reproducibility is another hallmark of RapiZyme Trypsin, achieved through its chemically optimized, recombinant (non-animal derived) formulation. Unlike traditional trypsin, RapiZyme is free from contaminating chymotrypsin and does not require treatment with TPCK inhibitors, which often introduce non-specific digestion challenges. The enzyme is lyophilized with an LC-MS compatible excipient, enhancing stability, activity, and vial recovery while ensuring predictable and consistent performance across batches and digestions. This reliability makes RapiZyme Trypsin an ideal choice for high-stakes applications where precision and reproducibility are non-negotiable.
