The RapiZyme Proteinase K Digestion Module comes ready to use and contains three easy-to-use solutions for efficient non-specific enzymatic degradation of proteins:
- RapiZyme Proteinase K has a sequence derived from T. album and is recombinantly expressed and purified from P. pastoris. Certified to be free of exonucleases, endonucleases, and ribonucleases, the solution ensures high quality, accurate, and precise nucleic acid measurements.
- As a subtilisin-type serine protease, RapiZyme Proteinase K exhibits optimal enzyme activity between 50–60 ℃, is active between pH 4 and 12, and is highly compatible with denaturants and surfactants such as 1M guanidine and 0.5% SDS.
- The Guanidine Tris Buffer Solution is a neutral pH solution of concentrated guanidine hydrochloride.
- The TCEP HCl concentrate is a stable solution format of a phosphine-based reducing agent solution.
Quick and Efficient Protein Digestion
The RapiZyme Proteinase K Digestion Module has been performance-tested for its ability to degrade plasma proteins in under 40 minutes. The accompanying chart shows an incubation time course for the digestion of plasma proteins at a 55 ℃ temperature in the presence of guanidine hydrochloride and TCEP reducing agent. SEC profiles are provided which demonstrate a shift of sample composition from large proteins to small peptides.
