Accurately, reproducibly, and easily perform protein conformation studies

The unprecedented system-level design integrates all the steps necessary to accurately, reproducibly, and most of all, easily, perform protein conformation studies. The system and its DynamX HDX Analysis Software remove the barriers to adopting high resolution LC and MS for routine studies of protein conformational changes, making hydrogen deuterium exchange with MS (HDX, HDX MS, HXMS, or H/D exchange) a core competency for your laboratory.

Higher efficiency separations

Carefully controlling temperature-sensitive parts of the HDX workflow ensures that the conformational changes are captured and not missed. UPLC separations of up to 15,000 psi take place within the cooled environment with the added advantage of sub-2-µm particles for enhanced chromatographic resolution and speed. Maintaining temperatures at 0±1 °C means that back-exchange is kept to an absolute minimum with the ACQUITY UPLC M-Class System with HDX Technology.

Once the HDX reaction has been chemically quenched and manually introduced, the HDX Manager directs the sample to the pepsin column for peptic digestion, or directly into the mass spectrometer (MS) for global analysis. The sample is then trapped for a rapid UPLC analytical separation.

ACQUITY UPLC M-Class System with HDX Technology FAQs

1. What is HDX technology?
Hydrogen-deuterium exchange (HDX) technology is a powerful technique used to study protein structure, dynamics, and interactions. In HDX, proteins are exposed to deuterium-containing solutions, allowing hydrogen atoms in the protein to exchange with deuterium. This exchange provides insights into protein conformations and binding sites by revealing regions of structural flexibility and stability.

The ACQUITY UPLC M-Class System is optimized for HDX workflows, offering precise temperature control, minimized back-exchange, and ultra-low dispersion to preserve sample integrity. When paired with mass spectrometry, the system enables high-resolution analysis of labeled peptides, making it ideal for structural biology studies.

2. What flow rate ranges does the system support?
The ACQUITY UPLC M-Class System is specifically designed for microflow separations, supporting flow rates typically in the range of 1–100 µL/min. These low flow rates enhance sensitivity by improving ionization efficiency and reducing solvent consumption, making the system well-suited for HDX experiments and other applications requiring precise control.

The system"s advanced fluidics ensure consistent performance at these low flow rates, delivering high resolution and reproducibility in chromatographic separations.
